The extracellular digestive apparatus of bivalve molluscs consists of a crystalline style containing diverse degradative enzymes which are released during rotation and dissolution of the style. This paper reviews the occurrence of a temperature-regulated bacteriolytic enzyme in the black mussel Choromytilus meridionalis , which is an inhabitant of coastal reefs on the south and south west coast of South Africa. This agent is unable to lyse Micrococcus luteus or Escherichia coli , both of which are susceptible to lysis by a true lysozyme. Abstract The changes in concentration of vitamin C, enzymatic and antioxidant activity during the ripening of two soursop selections (G1 and G2) at room temperature (22 ºC) and refrigeration (15 ºC) with an HR 85% were evaluated. The content of soluble protein, the activity of catalase (CAT), superoxide dismutase (SOD), peroxidase (POD), the concentration of vitamin C, as well as the antioxidant activity was evaluated by DPPH, ABTS and FRAP methods. The initial soluble protein concentration of the G1 and G2 selections diminished at 22 and 15 °C during ripening. Fruits stored at 22 °C showed the highest CAT activity. There are few studies about the impact of sewage sludge on the biological properties in Alfisols of the Chilean Coastal Range drylands. Hence, the objective of this study was to evaluate its effect on the microbial respiration and enzymatic activities of a degraded Alfisol located in the Bío Bío Region (Chile) that was cropped with yellow serradela (Ornithopus compressus L.). Characterization of a leucine aminopeptidase from Toxoplasma gondii OAK

The M17 family leucine aminopeptidase (LAP) hydrolyze amino acids from the N-terminus of peptides. Many LAPs from parasitic protozoa including Plasmmodium, Trypanosoma and Leishmania, have been intensely investigated because of their crucial roles in parasite biology. In this study, a functional recombinant Toxoplasma gondii LAP (rTgLAP) was expressed in Escherichia coli and its enzymatic activity against synthetic substrates for aminopeptidase, as well as the cellular localization was determined. Our results indicated that TgLAP is a functional aminopeptidase in the cytoplasma of T. gondii.

Alcohol dehydrogenases (ADH) are a class of enzymes that catalyze the reversible oxidation of alcohols to corresponding aldehydes or ketones, by using either nicotinamide adenine dinucleotide (NAD) or nicotinamide adenine dinucleotide phosphate (NADP), as coenzymes. In this study, a short-chain ADH gene was identified in Bombyx mori by 5'-RACE PCR. This is the first time the coding region of BmADH has been cloned, expressed, purified and then characterized. This study aimed to obtain the condition of maximum bromalein activity in different parts of pineapple plants produced in vitro, by micropropagation. The sStems and leaves of Pérola and Imperial cultivar plants were evaluated after three and eight months of in vitro cultivation in Murashige and Skoog medium without growth phytoregulator, macerated in potassium phosphate buffer at different pH values (5.7, 6.7 and 7.7). Total protein and proteolytic activity were determined in the samples after three- and eight-month cultivation periods. Knowledge of these enzymatic activities can help us to understand the species' digestive processes. The species chosen for this study were Ctenopharyngodon idella (herbivore), Rhamdia quelen (omnivore), Leporinus obtusidens (omnivore) and Hoplias malabaricus (carnivore). Dynamique et enzymologie de la digestion chez l'huitre Crassostrea gigas ArchiMer

Feeding experiments carried out on Crassostrea gigas allowed the authors to set up the stages and the timing of the movement of food throughout the digestive tract. Various activities involved in digestion (glycosidases, esterases, and peptidases) were investigated histochemically in the digestive tract in order to localize the sites of absorption and the sites of protein and carbohydrate digestion. An explicative model of the different stages of digestion is proposed.

Effect of broiler breeder age on pancreas enzymes activity and digestive tract weight of embryos and chicks Rev. Bras. Ciênc. Avic.

A study was carried out to evaluate the effect of broiler breeder age on the development of the digestive tract of embryos and chicks. Fertilized eggs Cobb from 30 and 60 week-old broiler breeder was utilized in this experiment. The results showed that eggs from older (60 weeks of age) broiler breeders were heavier (p = 0.001) than those from younger (30 weeks of age) broiler breeder. In addition, older broiler breeder had larger (p = 0.001) embryos showing a higher yolk sac (p = 0.001) and higher gastrointestinal tract relative weight (p = 0.007) than those from younger broiler breeder. The activity of three commercial microbial phytase (Aspergillus oryzae, A. niger, and Saccharomyces cerevisae) products used in broiler nutrition was determined at different pH (2.0 to 9.0) and temperature (20 to 90°C) values. Enzymatic activity was determined according to the reaction of the phytase with its substrate (sodium phytate), in four replicates, and was expressed in units of phytase activity (FTU). A. oryzae phytase exhibited optimal activity at pH 4.0 and 40°C, but its absolute activity was the lowest of the three phytases evaluated. Leporinus friderici is a South American promising species for fish culture. In French Guiana the "morphological species", L. friderici, L. granti, L. lebaili and L. affinis steyermarki are found. Their identification is sometimes difficult on usual criterion. To constitute an initial broodstock of L. friderici from local wild populations, the genetic structure of these species was studied. Fifteen enzyme systems, representing twenty loci, were screened.

Different parameters have been followed regularly at various times of anchovy (Engraulis encrasicholus ) ripening. Concentration in NaCl and water content as well as values of water activity and pH are constant and remain quite stable during the entire study period. Isolation and purification of bromelain from waste peel of pineapple for therapeutic application BABT

The aim of this work was to isolate and purify bromelain extracted from the pineapple peel by ammonium sulfate precipitation (40-80%), followed by desalting and freeze-drying with a 75% activity recovery and 2.2 fold increased specific activity. Ion exchange chromatography on DEAE-Sepharose was able to separate the polysaccharides from the enzyme, which was recovered in the elution step, maintaining its enzymatic activity. The batch adsorption of bromelain was evaluated in terms of total protein and enzymatic activity using Langmuir and Langmuir-Freundlich models. Results showed that the process could be suitable for the recovery and purification of the enzyme, maintaining its specific activity.

Microbial extracellular enzyme detection on agar plates by means of fluorogenic methylum-belliferyl-substrates. ArchiMer

A rapid and sensitive method to detect the extracellular enzymatic activity of bacteria colonies grown on agar plates is described. Selective agar media supplemented with protein, starch, chitin, Tween-80, etc. are conventionally used to detect biochemical properties of bacteria. It has been experimentally demonstrated with bacteria pure cultures that fluorogenic Methylumbelliferyl (MUF)-substrates are excellent substrate analogues for normally occurring polymers. Based on MUF-substrate hydrolysis the new method provides reliable qualitative estimates of extracellular enzymatic properties of bacteria within minutes using pure cultures as well as agar plates prepared for colony counts.

The influence of different commercial surfactants on the enzymatic activity of a commercial alpha-amylase from Bacillus licheniformis (Termamyl 300 L) has been studied. As non-ionic surfactants, alkyl polyglycosides (Glucopon® 215, Glucopon® 600 and Glucopon® 650) were studied, as were fatty alcohol ethoxylates (Findet 1214N/23 and Findet 10/15), and nonyl phenol ethoxylate (Findet 9Q/21.5NF).
